Shanks probably knows pluton is in wano since oden was with roger and told ben beckman "Let's get the one piece" as soon as kaido was defeated. Blackbeard knows of poseidon and pluton because of caribou and told catarina devon to get saturn's body (the ministry of science and defence) so that they could access the weapons of the holy land which might include uranus. Shanks is also keeping an eye on every movement of blackbeard so he knows which weapon he is gonna target first. Maybe shanks heard bits of it thru roger and had some inkling throughout their journey, but I think he heard most of it thru blackbeard during their fight. It was only then he decided that he needs to get more information by using his own origins to infiltrate. He knew that roger was just too early, so he bid his time until luffy. When he saw signs that he might be what joyboy was waiting for, he bet on luffy thus letting his arm cut off in order to sever his link with imu and mariejoia.
